Resumo: Indústrias de diferentes setores produzem efluentes em seus processos produtivos. Dependendo da atividade, as características dos resíduos produzidos variam consideravelmente. No caso da indústria sorveteira, esta apresenta efluentes de elevada carga orgânica proveniente, principalmente, do leite que é sua principal matéria-prima, refletindo, assim, em um resíduo com elevada Demanda Química de Oxigênio (DQO), óleos e graxas. Porém, o acúmulo de gorduras no sistema afeta a eficiência do tratamento do efluente gerado. Como forma de eliminar tais dificuldades, estuda-se a utilização de biocatalisadores como enzimas, em especial as lipases, que atuam na hidrólise de óleos e gorduras reduzindo, assim, a quantidade de lipídeos nos efluentes. Dessa forma, o presente estudo teve como objetivo produzir e aplicar uma lipase fúngica para degradação do efluente de uma indústria de sorvetes. Lipase do fungo endofítico Talaromyces sp., isolado de um hospedeiro amazônico, foi produzida em cultivo submerso e utilizada na degradação do efluente. A enzima foi produzida em Erlenmeyers com 100 mL de meio contendo azeite de oliva (1%) e pH 6. A atividade lipolítica máxima obtida foi de 3,192 ± 0,029 U/mL, após 144 h de cultivo. Este extrato enzimático foi aplicado diretamente no efluente autoclavado e no efluente bruto (sem autoclavagem), sendo a eficácia da hidrolise da gordura avaliada com base no índice de acidez (I.A) e na DQO. Verificou-se diminuição da DQO e aumento do I.A depois de 4 h. O tratamento enzimático com o efluente na forma bruta foi estudado utilizando-se um delineamento fatorial completo, com duas variáveis (pH e concentração enzimática) e dois níveis (pH = 3,5 e 7,0; concentração enzimática = 12,8 e 25,5 U/mL), durante 4 h. Os experimentos foram feitos avaliando-se a remoção de DQO e o aumento no I.A. Através dos gráficos de Pareto (p<0,05), verificou-se a influência estatisticamente significativa do pH sobre o I.A. Utilizando pH 7,0 e 12,8 U/mL de lipase foi possível obter os melhores resultados de degradação do efluente (88,46% de remoção de DQO e 22,22% de aumento no I.A.). O teor de óleos e gorduras (TOG) foi determinado tanto no efluente bruto, quanto naqueles experimentos realizados em pH 7,0, nas duas concentrações enzimáticas. A redução do TOG foi de 88,72% (com 12,8 U/mL) e de 94,50% (com 25,5 U/mL). Os resultados deste trabalho, além de mostrarem as variáveis que interferem na degradação de efluentes da indústria de sorvete, justificam a continuação de estudos de tratamento de efluentes com elevado teor lipídico, por meio da aplicação de lipase fúngica.
Abstract: Different types of industries generates effluents in their processes. Depending on the activity, the characteristics of the wastewater produced vary considerably. The ice cream industry has high organic load wastewater from milk, which is its main raw material, reflecting in a residue with high Chemical Oxygen Demand (COD) and high content of oils and greases. However, the accumulation of fats in the system affects the efficiency of treatment of the generated effluent. In a way to eliminate such difficulties, the use of biocatalysts as enzymes is studied, especially lipases, which act in the hydrolysis of oils and fats, thus reducing the amount of lipids in the effluents. Thus, the present study aimed to produce and apply a fungal lipase for effluent degradation of an ice cream industry. Lipase of the endophytic fungus Talaromyces sp., Isolated from an Amazonian host, was produced in submerged cultivation and used for effluent degradation. The enzyme was produced in Erlenmeyers with 100 mL medium containing 1% olive oil and pH 6. The maximum lipolytic activity obtained was 3.192 ± 0.029 U / mL after 144 h of cultivation. This enzymatic extract was applied directly to the autoclaved effluent and the raw effluent (without autoclaving). The effectiveness of fat hydrolysis was evaluated based on the acidity index (A.I) and COD. There was a decrease in COD and an increase in A.I after 4 h. Enzymatic treatment with raw effluent was studied using a complete factorial design with two variables (pH and enzymatic concentration) and two levels (pH = 3.5 and 7.0; enzymatic concentration = 12.8 and 25.50 U/ml) for 4 h. The experiments were performed by evaluating COD removal and increase in A.I. Through Pareto graphs (p <0.05) was verified the statistic influence of pH on the A.I. Using pH 7.0 and 12.8 U/mL lipase, it was possible to obtain the best effluent degradation results (88.46% COD removal and 22.22 % increase in A.I.). The oil and fat content (TOG) was determined both in the raw effluent and in those experiments performed at pH 7.0, in both enzymatic concentrations. The TOG reduction was 88.72 % (with 12.8 U / mL) and 94.50 % (with 25.5 U/mL). The results of this work, besides showing the variables that interfere with the effluent degradation of the ice cream industry, justify the studies of high lipid effluent treatment through the application of fungal lipase.
