Título: Estudo químico e biológico de Casearia javitensis Kunth

Resumo: No Brasil, especialmente na região Amazônica, existe uma grande biodiversidade de espécies vegetais que ainda não foram estudadas química e biologicamente. Nesse sentido, o presente trabalho teve como objetivo realizar o estudo químico e biológico dos extratos de Casearia javitensis Kunth. Assim os extratos hexânicos e metanólicos bem como as fases obtidas dos extratos metanólicos dos galhos e das folhas de Casearia javitensis foram testados para determinar seu potencial antibacteriano, mediante o método de difusão em poço, também para verificar a atividade antioxidante (DPPH e Fe3+/fenantrolina), e de toxicidade frente à Artemia salina. As frações obtidas da fase acetato de etila dos extratos metanólicos das folhas e dos galhos apresentaram um alto potencial antioxidante, sendo que a análise em CCDC dessa fração foi positiva com FeCl3, indicando a possível presença de substâncias fenólicas. Os extratos hexânicos dos galhos e das folhas apresentaram atividade frente às bactérias Corynebacterium glutamicum, Staphylococcus aureus e Bacillus cereus. Os extratos metanólicos de folhas e galhos foram ativos frente às bactérias Pseudomonas aeruginosa, Staphylococcus aureus e Serratia marcescens. Os ensaios frente à A. salina mostraram que nenhum dos extratos foram considerados como tóxicos. O fracionamento dos extratos hexânicos e metanólicos das folhas permitiu o isolamento de duas substâncias: o -sitosterol e o 4-hidroxifenil-6-cafeoil-β-L-glicosídeo (um fenol-glicosídeo), respectivamente, as quais foram identificadas através da análise dos espectros de RMN de 1H e 13C e de espectrometria de massas. Abstract: In Brazil, especially in the Amazon region, there is a great biodiversity of plant species that have not been studied chemical and biologically yet. Therefore the aim of the present study was to perform chemical and biological studies of Casearia javitensis Kunth extracts. Then the hexanic and methanolic extracts as the fractions obtained from the methanolic extract of the branches and leaves from Casearia javitensis have been tested in order to identify the antimicrobial activity by diffusion method well, antioxidant activity (DPPH and Fe3+/phenanthroline) and toxicity against Artemia salina. The fractions of ethyl acetate phase obtained from the methanol extracts of the leaves and branches showed a high antioxidant potential, and the analysis by TLC of these fractions were revealed with FeCl3, indicating the presence of phenolic compounds. The hexanic extract of the branches and leaves showed activity against the bacteria: Corynebacterium glutamicum, Staphylococcus aureus and Bacillus cereus. The methanolic extracts of leaves and branches were active against Pseudomonas aeruginosa, Staphylococcus aureus and Serratia marcescens. The assay against A. salina showed that none of these extracts were considered toxic. The fractionation of the hexane and methanolic extracts of the leaves yielded two compounds: -sitosterol and 4hydroxyphenyl-6-caffeoyl-β-L-glycoside (a phenol-glycoside), respectively, which were identified by 1H and 13C NMR analysis and mass spectrometry.
